以萌发初期大豆弯曲的子叶节为靶点,利用草甘膦(Roundup)点施鉴定法,分析了221份大豆微核心种质的草甘膦耐受性及其与抗草甘膦转CP4-EPSPS基因大豆AG5601的差异,结果表明,草甘膦对不同大豆种质的抑制程度与点施的草甘膦浓度呈正相关,Roundup稀释浓度为1/1000、1/10000时,草甘膦对大豆生长几乎无影响;Roundup稀释浓度为1/10时,草甘膦显著抑制大豆生长,导致植株死亡;Roundup稀释浓度为1/100时,不同大豆种质对草甘膦耐受性差异显著,并鉴定出对草甘膦具有较好耐受性的种质10份。虽然大豆微核心种质对草甘膦的耐受程度远远低于AG5601,但不同大豆种质对草甘膦耐受性存在显著差异,这为利用转基因和杂交转育技术培育抗草甘膦转基因大豆的受体或轮回亲本的选择提供了理论依据。 相似文献

A novel ribosome-inactivating protein designated Moschatin from the mature seeds of pumpkin (Cucurbita moschata) has been successively purified to homogeneity, using ammonium sulfate precipitation, CM-cellulose 52 column chromatography, Blue Sepharose CL-6B Affinity column chromatography and FPLC size-exclusion column chromatography. Moschatin is a type 1 RIP with a pI of 9.4 and molecular weight of~29 kD. It is a rRNA N-glycosidase and potently blocked the protein synthesis in the rabbit reticulocyte lysate with a IC_(50) of 0.26 nM. Using the anti-human melanoma McAb Ng76, a novel immunotoxin Moschatin-Ng76 was prepared successfully and it efficiently inhibited the growth of targeted melanoma cells M_(21) with a IC_(50) of 0.04 nM, 1500 times lower than that of free Moschatin. The genome of a new SV40 strain (SV-IMB) isolated from a rhesus monkey was completely sequenced and compared with other isolates. The results showed that the whole genome contains 5246bp, and the average identity of SV-IMB was 98.1% as compared to other SV40 isolates. Its regulatory region is composed of a complete enhancer and a defective enhancer. Amino acid changes occurred to some extent in both the large T antigen (T-Ag) and VP1 region. The St and E are two important basic genomes in the perennial tribe Triticeae (Poaceae). They exist in many perennialspecies and are very closely related to the A, B and D genomes of bread wheat (Triticum aestivum L.). Genomic Southernhybridization and genomic in situ hybridization (GISH) were used to analyze the genomic relationships between the twogenomes (St and E) and the three basic genomes (A, B and D) of T. aestivum. The semi-quantitative analysis of the Southernhybridization suggested that both St and E genomes are most closely related to the D genome, then the A genome, andrelatively distant to the B genome. GISH analysis using St and E genomic DNA as probes further confirmed the conclusion.St and E are the two basic genomes of Thinopyrum ponticum (StStE~eE~bE~x) and Th. intermedium (StE~eE~b), two perennialspecies successfully used in wheat improvement. Therefore, this paper provides a possible answer as to why most of thespontaneous wheat-Thinopyrum translocations and substitutions usually happen in the D genome, some in the A genomeand rarely in the B genome. This would develop further use of alien species for wheat improvement, especially thosecontaining St or E in their genome components. 相似文献
